Yes, the culture can be pretty different from state to state and the two states you mentioned are pretty much at opposite ends of the American cultural spectrum. Massachusetts is one of the most progressive US states (it's the first state to legalize gay marriage for example) and among the least religious, while Kansas is fairly conservative and very religious (they actually teach "intelligent design" in the public schools there). Also Massachusetts has one of the highest population densities in the country and Kansas is largely farm country so there's a big urban versus rural contrast there as well. As a result the cultures of those two states are quite different.
